A petri dish is a conventional laboratory vessel, consisting of a flat disc-shaped bottom and a cover, mainly made of plastic and glass, and glass can be used for plant materials, microbial culture and animal cell adherent culture. Most of the plastic is disposable, suitable for laboratory inoculation, streaking, and isolation of bacteria for the cultivation of plant materials.
Method/step:
1
Petri dishes are usually made of solid medium for plate culture (that is the origin of the name of the plate plate). The production of plate medium is to dissolve the installed sterilized agar medium with warm water (sterile), remove the test tube cotton plug, Pass the mouth of the tube over the flame of the alcohol lamp, and then slightly open the lid of the sterilized culture dish, so that the mouth of the test tube can go deep. It is evenly distributed on the bottom of the dish and condensed to obtain a plate culture medium.
2
Because the reproduction, development and growth of bacteria are directly related to the supplied medium (nutrition), especially for quantitative inspection and analysis, it has decisive significance for the amount of nutrients provided.
3
The amount of nutrition provided during bacterial culture, whether it is uniform, and whether the bottom of the petri dish is flat are extremely important. If the bottom of the petri dish is uneven, the distribution of the agar medium will vary depending on whether the bottom of the petri dish is flat or not. The supply is insufficient, which is closely related to quantitative analysis, so the bottom of the quantitative petri dish is required to be particularly flat due to the reason. However, for general characterization (inspection of bacteria, colony growth, reproduction, etc.), ordinary petri dishes can be used.
Precautions
After cleaning and disinfection before use, whether the petri dish is clean or not has a great influence on the work, which can affect the pH of the medium. If there are certain chemicals, it will inhibit the growth of bacteria.
